Mechanism of action
Daunorubicin is an anthracycline antibiotic active in all phases of the cell cycle with maximal
activity in S phase 2. It is an analogue of Doxorubicin. It has several modes of action
including intercalation between DNA base pairs, inhibition of DNA and RNA synthesis by
template disordering and steric obstruction 2, topoisomerase II mediated DNA damage,
production of oxygen- free radicals which cause damage to DNA and cell membranes 3,4.
Metal daunorubicin complexes, particularly with iron, may contribute to cardiotoxicity by
toxic free radical generation 3,4.
