Mechanism of action:
Dactinomycin is an antineoplastic antibiotic. Mechanism of cytotoxicity includes
intercalation with guanine residue of DNA thereby forming a complex with DNA. This
impairs the template activity of DNA and therefore inhibits DNA-dependent RNA synthesis.
Protein and DNA synthesis are also inhibited but at higher concentrations of dactinomycin1.
Considerations prior to chemotherapy
• Ensure good robust venous access
• Full blood count
• Liver function tests2
• Creatinine, urea and electrolytes
